ABSTRACT

Objective To understand the influencing factors of stroke symptoms in middle-aged and elderly patients with hypertension in the community, and to provide reference for the prevention and control of stroke. Methods A total of 410 cases of middle-aged and elderly hypertensive patients registered in community health service centers (or health centers) were randomly selected from 2 administrative regions of Wuhan to conduct a unified questionnaire survey. Chi-square test, analysis of variance and multivariate logistic regression were used to analyze the influencing factors of stroke symptoms in hypertensive patients. Results Of 31.83% the study subjects had stroke symptoms. The analysis of the results showed that high monthly per capital household income, adequate fruit intake and high level of dietary knowledge were protective factors for stroke symptoms in middle-aged and elderly patients with hypertension. Hypertension complications, annual medical check-ups in the hospital and a history of stroke / heart disease in immediate family members were risk factors for stroke symptoms. Conclusion Improving the health awareness of middle-aged and elderly people, vigorously promoting health education and raising the level of social security for low-income groups can reduce the risk of stroke in middle-aged and elderly patients with hypertension.

ABSTRACT

Objective:To establish and validate the prognosis risk scoring system based on the prognostic risk factors of esophageal squamous cell carcinoma (ESCC).Methods:The clinical data of 135 patients with ESCC in Rugao Hospital Affiliated to Nantong University from January 2014 to December 2017 were retrospectively analyzed. All patients underwent radical resection of esophageal cancer and were followed up until February 1, 2020. According to prognosis, the patients were divided into survival group (70 cases) and death group (65 cases). Cox proportional hazard model was used to analyze the risk factors of affecting prognosis, and the prognostic risk scoring system was established according to the HR value. The patients were divided into low, medium and high risk of death, and the prognosis of patients with different death risk was compared.Results:The median follow-up time was 41 (3 to 72) months and the median survival time was 51 months. Lymphocyte to monocyte ratio (LMR), prognostic nutritional index (PNI) and C-reactive protein (CRP) were independent risk factors for the prognosis of ESCC patients ( P < 0.05). The 1-, 3- and 5-year cumulative survival rates of patients with low risk of death were 96.7%, 77.7% and 65.2%, respectively, which were higher than those of patients with moderate risk of death (91.7%, 75.6% and 37.1%) and those of patients with high risk of death (87.9%, 42.2% and 26.8%), and the difference was statistically significant ( χ2 = 18.462, P < 0.01). Conclusions:The prognosis risk scoring system is an effective evaluation system for the prognosis of ESCC patients, which can help clinicians to stratify patients and perform individualized treatment.

ABSTRACT

@#【Objective】 To investigate the predictive value of preoperative Gd- EOB- DTPA enhanced MRI in the expression of cytokeratin 19(CK19)in hepatocellular carcinoma(HCC).【Methods】A total of 102 patients,including 94 male and 8 female,with single HCC confirmed by pathology after operation who underwent preoperative enhanced MRI were retrospectively analyzed. A total of 25 were CK19-positive HCC and 77 were CK19-negative HCC. Two radiologists evaluated MR features including tumor size,tumor margin,intratumoral vessels,signal intensity(SI)on arterial phase (AP) ,enhancement pattern ,arterial rim enhancement ,peritumoral enhancement ,internal cystic or necrotic portion,hemorrhage,intratumoral fat,tumor capsule,vascular invasion,lymph node metastasis,intratumoral septum, target sign on diffusion weighted imaging(DWI)or hepatobiliary phase(HBP),peritumor hypointensity,SI on ADC,SI on HBP ,T1 relaxation times and T1 reduction rate between pre- and post- contrast enhancement. The associations between these imaging features and CK19 expression were investigated. 【Results】SI on AP(P = 0.013),arterial rim enhancement(P = 0.018),target sign on DWI(P = 0.001)and target sign on HBP(P = 0.005)were significantly associated with CK19 expression. Delayed enhanced intratumoral septum(P = 0.042)was associated with CK19 expression between HCCs less than 5 cm. Target sign on DWI(P = 0.001,OR = 4.875,95%CI:1.838~12.927)were independent significant factors of CK19- positive HCC.【Conclusion】Preoperative enhanced MRI with Gd- EOB- DTPA is helpful to predict CK19 expression of HCC.

ABSTRACT

OBJECTIVE: To study the effects of Tiaopi huxin prescription (TPHXP) on the atherosclerosis (AS) of ApoE-/- mice, and to investigate its mechanism. METHODS: Forty male ApoE-/- mice were divided into blank group, model group, simvastatin group (positive control, 5 mg/kg) and TPHXP low-dose and high-dose groups (50, 150 mg/kg), with 8 mice in each group. Except that blank group was given common diet, other groups were given high-lipid diet to induce AS model. After modeling, administration groups were given relevant medicine intragastrically, and blank group and model group were given constant volume of normal saline intragastrically, once a day, for consecutive 12 weeks. After last medication, the serum levels of TC, TG, LDL-C and HDL-C were determined by spectrophotometry. The serum level of NO was detected by nitrate reduction method. The serum levels of IL-6 and VCAM-1 were determined by ELISA. After separating thoracic aorta, HE staining was used to observe the formation of plaque in the thoracic aorta of mice in each group, and the corrected plaque area was calculated. Western blotting was conducted to determine the expression of NF-κB p65, Cav-1 and eNOS. RESULTS: Compared with blank group, the serum levels of TC, TG, LDL-C, IL-6 and VCAM-1 were increased significantly in model group, while the levels of HDL-C and NO were decreased significantly (P<0.01). The plaque of thoracic aorta was obvious and the corrected plaque area were increased significantly (P<0.01). The relative expression of NF-κB p65 and Cav-1 were increased significantly, while the relative expression of eNOS was decreased significantly (P<0.01). Compared with model group, the serum levels of TC, TG and LDL-C in administration groups, the serum levels of IL-6 and VCAM-1 in simvastatin group and TPHXP high-dose group were decreased significantly, while the serum levels of HDL-C and NO were increased significantly in administration groups (P<0.05 or P<0.01). In administration groups, the plaques of thoracic aorta were reduced and the corrected plaque area was decreased significantly (P<0.05 or P<0.01); the relative expression of NF-κB p65 and Cav-1 were decreased significantly, while the relative expression of eNOS was increased significantly (P<0.05 or P<0.01). CONCLUSIONS: TPHXP can regulate the level of blood lipid, decrease the level of inflammatory factors and inhibit the formation of AS plaque, the mechanism of which may be associated with inhibiting Cav-1/NF-κB pathway.

ABSTRACT

Objective To investigate the effect of erythropoietin(EPO)on the proliferation of neural stem cells(NSCs)derived from central canal of adult rat spinal cord in vitro ,so as to provide a theoretical basis for clinical treatment of spinal cord injury by autotransplanting or allograft transplanting of adult spinal cord NSCs. Method NSCs were isolated from the central canal of the adult rats spinal cord by microsurgical method,and Nestin(nestin)and Sox2 immunofluorescence stain were used to identify the cells. After cells were treated with different dose of EPO,5,10,20 and 40 U/mL,respectively,the optical treatment concentration and time were determined by CCK8 assay. The effect of EPO on the cell count and the expression of Cyclin D1 in NSCs were detected at the treatment time 96 h. Result The NSCs derived from the central canal of adult SD rats spinal cord could stably express protein Nestin and transcription factor Sox2. As the results of CCK8 test,cell counts and real-time quantitative PCR showed the optimal treatment of concentration and time maybe 20 U/mL and 96 h. Conclusions This study shows that EPO can promote the proliferation of NSCs derived from central canal of adult rat spinal cord,and the optimal treatment of concentration and time for proliferation might be 20 U/mL and 96 h.

ABSTRACT

Objective To investigate the effect of amentoflavone(AMF)on inhibiting wear debris-in-duced osteolysis in vivo and RANKL-induced osteoclastogenesis in vitro. Methods Twenty-four male C57BL/J6 mice were randomly divided into four groups:the control group,titanium group,the low and high concentration group.Micro-CT and histological analysis were performed.CCK-8 assay was used to determin the effect of AMF on the proliferation of BMMs.TRAP staining and bone resorption assays were used to investigate the effect of AMF on osteoclastogenesis and function. The effect of AMF on prevention MAPK signaling was detected by Western blot assay. Results AMF could prevent osteolysis in vivo and suppress osteoclastogenesis,bone resorption without cytotoxicity in vitro.AMF suppressed MAPK signaling pathways.Conclusion AMF could inhibit osteoclastogenesis and titanium debris-induced osteolysis through suppressing MAPK signaling.
